Product Description
To obtain good preparations of eukaryotic mRNA, it is necessary to minimize the activity of RNase liberated during cell lysis from the cells and to protect the contamination of RNase from outside during the overall process. Guanidine HCl and guanidinium thiocyanate can denature proteins involving cell composition and RNase. The contamination of RNase from outside of tubes should be blocked for obtaining intact RNAs. Diethyl pyrocarbonate (DEPC) is widely used to inactivate foreign RNase in solution, glassware, plasticware, etc. DEPC is the modification reagent for histidine and tyrosine residues in proteins, thus can denature proteins.
ML 019-01 is prepared as follows. 1 mL of DEPC is added to 1 L of Ultra Pure Water (ML 019-02) and dissolved completely for over than 12 hours. DEPC is removed by autoclaving (121°C, 15 lb/sq. in., 20 min). ML 019-01 is used for RNase free solution and for preparation of glasswares or plasticwares.
Storage/Stability
RNase free water should be stored at 15~30°C. Deterioration of the solution may be recognized by (1) precipitate or particulate matter throughout the solution, (2) cloudy appearance, (3) color change, and/or (4) pH change. Product label bears expiration date.
