Product Description
Buffered saline (BS) is one of the balanced salt solutions bearing the buffering capacity and capable of maintaining osmotic balance similar to the natural condition of body. Of many salines, phosphate-buffered saline (PBS), Dulbecco’s phosphate-buffered saline (D-PBS), Earle’s balanced salt solution (EBSS), and Hanks’ balanced salt solution (HBSS) are widely used because the buffering capacity of phosphate is effective at in vivo neutral pH with major roles in maintaining viability or integrity rather than promoting growth or functions of cells or molecules. Among these BSs, PBS and D-PBS contain the basic inorganic salts. Other principal functions of BSs are as follows; (1) dissolve or dilute materials for use in cell culture, (2) provide inorganic salts essential for the basic cell metabolism, (3) maintain the osmotic balance, (4) maintain pH with buffering capacity, and (5) eliminate certain components as serum or trypsin.
Storage/Stability
The buffered salines should be stored at 15~30°C. For long time storage, store at 2~8°C. Deterioration of the liquid may be recognized by (1) precipitate or particulate matter throughout the solution, (2) cloudy appearance, (3) color change, and/or (4) pH change. The nature of supplements added may affect storage conditions and shelf life of the liquid. Product label bears expiration date.
Biological Performance Characteristics
The growth-maintaining capacity of EBSS is tested in an appropriate medium after storage of an appropriate cell line. Growth rates are examined through three subculture generations and compared with parallel cultures using the standardized control saline. Cells are counted and growth is plotted as a logarithmic function of time in culture, and seeding efficiency, doubling time, and final cell density are determined. During the testing period cultures are examined microscopically for a typical morphology and evidence of cytotoxicity.
